Definition: Technique of DNA analysis using electrophoretic methods to separate large DNA framents, up to 2 million base pairs in size, produced by digesting DNA with restriction enzymes with relatively long DNA recognition sequences which as a consequence cut the DNA relatively infrequently.
Context: Use of pulsed field gel electrophoresis (PFGE), in which DNA fragments are subjected alternately to two approximately perpendicular fields, enables separation of DNA fragments of up to 2 000 000 base-pairs in size and with recent improvements in the technique up to 5–10 000 000 base-pairs in size.
